Diversity of Endophytic Microbes in Taxus yunnanensis and Their Potential for Plant Growth Promotion and Taxane Accumulation

Taxus spp. are ancient tree species that have survived from the Quaternary glacier period, and their metabolites, such as taxol, been used anticancer drugs globally. Plant–endophytic microbial interaction plays a crucial role in exerting profound impact on host growth secondary metabolite synthesis. In this study, high-throughput sequencing was employed to explore endophytic diversity roots, stems, leaves of yunnanensis (T. yunnanensis). The analysis revealed some dominant genera bacteria, Pseudomonas, Neorhizobium, Acidovorax, Flavobacterium, with Cladosporium, Phyllosticta, Fusarium, Codinaeopsis prominent fungi genera. We isolated 108 bacteria 27 leaves. vitro assays were utilized screen for growth-promoting capabilities, including IAA production, cellulase, siderophore protease ACC deaminase activity, inorganic phosphate solubilization, nitrogen fixation. Three promising strains, Kocuria sp. TRI2-1, Micromonospora TSI4-1, Sphingomonas MG-2, selected based superior growth-promotion characteristics. These strains exhibited preferable plant promotion when applied Arabidopsis thaliana growth. Fermentation broths these three also found significantly promote accumulation taxanes T. stem cells, among which strain TSI4-1 demonstrated outstanding increase potentials, an effective induction baccatin III, 10-DAB contents. After six days treatment, contents metabolites 3.28 times, 2.23 2.17 times initial amounts, reaching 8720, 331, 371 ng/g dry weight respectively. findings present new insight into industrialization taxol production through cell fermentation, thereby promoting conservation wild resources by maximizing potential economic benefits.
